研究加氏乳杆菌TF08-1对葡聚糖硫酸钠(DSS)诱导的溃疡性结肠炎(UC)小鼠模型的影响及机制。
方法

将24只8周龄C57BL/6N雄鼠随机分为4组,每组6只,分别为对照(Control)组、模型(DSS)组、美沙拉秦(5-ASA+DSS)组、加氏乳杆菌(TF08-1+DSS)组,采用30 g/L葡聚糖硫酸钠饮水饲喂的方法建立小鼠UC模型,期间记录体质量变化、疾病活动指数(DAI)评分,第8天处死后测量结肠长度,HE染色评估组织病理学变化,酶联免疫吸附测定(ELISA)检测肠组织IL-10、TNF-α表达量,Western blot检测Bcl-2表达量,TUNEL染色检测细胞凋亡。
结果

与模型组相比,加氏乳杆菌组DAI评分降低,结肠缩短程度较轻,HE染色显示加氏乳杆菌组结肠组织损伤减轻;加氏乳杆菌组结肠组织IL-10、TNF-α水平下降;Bcl-2蛋白表达量增加,TUNEL染色显示黏膜凋亡细胞显著减少[病理评分:7.17

加氏乳杆菌TF08-1可缓解DSS诱导的小鼠溃疡性结肠炎, 减轻炎症反应,其可能通过抑制肠黏膜凋亡发挥作用。
Objective

To explore the effects of Lactobacillus gasseri TF08-1 on ulcerative colitis (UC) mice induced by Dextran Sulfate Sodium Salt (DSS) and the related mechanisms.
Methods

Twenty-four 8-week-old C57BL/6N mice were randomly and equally divided into Control group, DSS group, 5-ASA+DSS group, and TF08-1+DSS group. UC model was established by feeding with 30 g/L dextran sodium sulfate drinking water for 7 days. During this period, 5-ASA was given to 5- ASA+DSS group and TF08-1 was given to TF08-1+DSS group. The weight change and the disease activity index (DAI) score were recorded every day. The mice were sacrificed after 7 days of administration, and the colon length was measured. HE staining was used to evaluate histopathology, Enzyme-linked immunosorbent assay (ELISA) to detect intestinal tissue IL-10 and TNF-α level, Western blot to detect Bcl-2 expression, and TUNEL staining to detect cell apoptosis.
Results

No death was observed in all of the mice. Compared with DSS group, the DAI score of the TF08-1+DSS group was lower, and the extent of colon shortening was lighter. HE staining showed that the colonic tissue damage of TF08-1+DSS group was reduced. The levels of IL-10 and TNF-α in the colonic tissue decreased in TF08-1+DSS group, while Bcl-2 protein expression increased. TUNEL staining showed a significant decrease in mucosal apoptotic cells [pathological score: 7.17

Conclusion

Lactobacillus gasseri TF08-1 alleviates DSS-induced ulcerative colitis in mice and reduces inflammation, which may play a role by inhibiting intestinal mucosal apoptosis.
