探索霍乱弧菌新发菌株H1的六型分泌系统(T6SS)存在的转录调控机制。
方法

本研究以转录组测序为基础,分析H1株分别由于
vipA
敲除(H1Δ
vipA
),
vipB
敲除(H1Δ
vipB
)和
clpV
敲除(H1Δ
clpV
)时T6SS基因簇的转录水平,并对其差异基因进行了基因本体论(GO)富集和京都基因与基因组百科全书(KEGG)富集。
结果

发现H1Δ
vipA
有22个差异基因(FDR
<
0.01,|log
2
FC|≥1),其中有18个属于T6SS基因簇;并且除
vipA
外,其转录水平均呈上升趋势。结合GO富集和KEGG富集结果,发现
vipA
的缺失会引起与霍乱弧菌群体感应相关基因(
hapR
)的转录水平显著降低。
结论

我们得出结论
vipA
基因在霍乱弧菌H1菌中是一个具有多重功能的基因,一是编码T6SS鞘结构蛋白,二是参与T6SS调控;推测
hapR
可能参与H1株中
vipA
对T6SS基因簇的转录调控。

To explore the transcriptional control mechanism of type Ⅵ secretion system (T6SS) in the new strain of
Vibrio cholerae
H1.
Methods

This study was based on transcriptome analyzed the transcription level of T6SS gene clusters of H1 strain separately knockout
vipA
(H1Δ
vipA
),
vipB
(H1Δ
vipB
) and
clpV
(H1Δ
clpV
), and carried out Gene ontology (GO) enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG) enrichment of the differential genes.
Results

It was found that there were 22 differential genes (FDR
<
0.01, |log2FC|≥1) in H1Δ
vipA
, 18 of which belong to the T6SS gene cluster, and besides
vipA
, the average transcribed level showed an upward trend. Combined with GO enrichment and KEGG enrichment results, it was found that the absence of
vipA
decreased the transcription levels of the quorum sensing gene (
hapR
).
Conclusions

We concluded that the
vipA
gene is a multi-functional gene in
Vibrio cholerae
H1, one is encoded T6SS structure protein and the other is taken apart in T6SS regulation,and we speculate
hapR
may have a role to play in the transcription regulation of T6SS gene clusters by
vipA
in H1 strain.
